Growing competition and economic recession is driving the need for more rapid redesign of operations enabled by innovative technologies. The acquisition, development and implementation of systems to manage customer complaints and control the quality assurance process is a critical area for engineering and manufacturing companies. Multimethodologies, and especially those that can bridge ‘soft’ and ‘hard’ OR practices, have been seen as a possible means to facilitate rapid problem structuring, the analysis of alternative process design and then the specification through to implementation of systems solutions. Despite the many ‘hard’ and ‘soft’ OR problem structuring and management methods available, there are relatively few detailed empirical research studies of how they can be combined and conducted in practice. This study examines how a multimethodology was developed, and used successfully, in an engineering company to address customer complaints/concerns, both strategically and operationally. The action research study examined and utilised emerging ‘soft’ OR theory to iteratively develop a new framework that encompasses problem structuring through to technology selection and adoption. This was based on combining Soft Systems Methodology (SSM) for problem exploration and structuring, learning theories and methods for problem diagnosis, and technology management for selecting between alternatives and implementing the solution. The results show that, through the use of action research and the development of a contextualised multimethodology, stakeholders within organisations can participate in the design of new systems and more rapidly adopt technology to address the operational problems of customer complaints in more systemic, innovative and informed ways.  相似文献

We consider the problem of optimizing superresolution microscopy with photoswitchable molecules that irreversibly photobleach. Using variational methods, we show that the number of single-molecule images is maximized in a simple scheme with a constant number of activated fluorophores per cycle. For high-speed acquisition, deviations from the optimal scheme do reduce the information collected but fortuitously also reduce certain types of errors, making the scheme very robust. Finally, we calculate the amount of information lost due to bleaching and noise.  相似文献

Enzymatic N(2) reduction proceeds along a reaction pathway composed of a sequence of intermediate states generated as a dinitrogen bound to the active-site iron-molybdenum cofactor (FeMo-co) of the nitrogenase MoFe protein undergoes six steps of hydrogenation (e(-)/H(+) delivery). There are two competing proposals for the reaction pathway, and they invoke different intermediates. In the 'Distal' (D) pathway, a single N of N(2) is hydrogenated in three steps until the first NH(3) is liberated, and then the remaining nitrido-N is hydrogenated three more times to yield the second NH(3). In the 'Alternating' (A) pathway, the two N's instead are hydrogenated alternately, with a hydrazine-bound intermediate formed after four steps of hydrogenation and the first NH(3) liberated only during the fifth step. A recent combination of X/Q-band EPR and (15)N, (1,2)H ENDOR measurements suggested that states trapped during turnover of the α-70(Ala)/α-195(Gln) MoFe protein with diazene or hydrazine as substrate correspond to a common intermediate (here denoted I) in which FeMo-co binds a substrate-derived [N(x)H(y)] moiety, and measurements reported here show that turnover with methyldiazene generates the same intermediate. In the present report we describe X/Q-band EPR and (14/15)N, (1,2)H ENDOR/HYSCORE/ESEEM measurements that characterize the N-atom(s) and proton(s) associated with this moiety. The experiments establish that turnover with N(2)H(2), CH(3)N(2)H, and N(2)H(4) in fact generates a common intermediate, I, and show that the N-N bond of substrate has been cleaved in I. Analysis of this finding leads us to conclude that nitrogenase reduces N(2)H(2), CH(3)N(2)H, and N(2)H(4) via a common A reaction pathway, and that the same is true for N(2) itself, with Fe ion(s) providing the site of reaction.  相似文献

Recently a new framework has been proposed to explore the dynamics of pseudoperiodic time series by constructing a complex network [J. Zhang, M. Small, Phys. Rev. Lett. 96 (2006) 238701]. Essentially, this is a transformation from the time domain to the network domain, which allows for the dynamics of the time series to be studied via organization of the network. In this paper, we focus on the deterministic chaotic Rössler time series and stochastic noisy periodic data that yield substantially different structures of networks. In particular, we test an extensive range of network topology statistics, which have not been discussed in previous work, but which are capable of providing a comprehensive statistical characterization of the dynamics from different angles. Our goal is to find out how they reflect and quantify different aspects of specific dynamics, and how they can be used to distinguish different dynamical regimes. For example, we find that the joint degree distribution appears to fundamentally characterize spatial organizations of cycles in phase space, and this is quantified via an assortativity coefficient. We applied network statistics to electrocardiograms of a healthy individual and an arrythmia patient. Such time series are typically pseudoperiodic, but are noisy and nonstationary and degrade traditional phase-space based methods. These time series are, however, better differentiated by our network-based statistics.  相似文献
